No, you don't need to delete anything from iTunes. Make a new playlist called iPhone, and drag a bunch of songs into it from your main library. Then when the phone is connected click the Music tab and tell it to sync to that playlist. Then everytime you sync it'll make sure the device is up to date with whatever is in the playlist. Means you can choose what goes on and off it when it's not even hooked up, which is useful.
You can sync more than 1 playlist if you want, I just use the one cos I have an 8gb 3G and I dedicate about 4gb to MP3, 2gb to podcasts and the rest to apps.
If you don't want to sync to a playlist, just click "Manually manage music and videos" on the summary tab.
